Intervertebral disc regeneration in a rat model via nanocomposite hydrogel loaded with palette rich plasma growth factors, endometrial stem cells, and melatonin: An in vitro and in vivo study
In the context of spinal health and musculoskeletal disorders, the challenge of intervertebral disc degeneration is a significant concern that impacts millions of individuals worldwide. In the current study, platelet rich plasma growth factors, endometrial stem cells, and melatonin-loaded chitosan nanoparticles were impregnated into a calcium alginate hydrogel in order to develop a filling material for treating intervertebral disk injury. The produced nanocomposite delivery system did not show significant toxicity against endometrial stem cells and protected them against oxidative stress. By the end of the 7th day, the cumulative release profile was quantified at 55.79 +/- 5.95%. In addition, our delivery system exhibited antioxidative activity and augmented the migration activity of stromal cells. In vivo study showed that the sophisticated delivery system significantly augmented intervertebral disk injury repair compared with other experimental groups. ELISA assay results showed that the treatment reduced the tissue concentrations of IL-6 and TNF-a and upregulated TGF-beta expression levels. Our developed system may have augmented intervertebral disk injury repair through modulation of inflammation.
